Abstract of talk:

Proteomics research is now at a breakneck speed after the completion of the human genome sequence. Cellular proteins confers the properties & functions of cells and is tightly regulated by genetic & epigenetic changes as the cell grows, divides, differentiates or de-differentiates as in the case of tumorigenesis.

Cellular protein detection is therefore an important technique in biological science. It ranges from western blotting, immunohistochemistry, immunocytochemistryIt immunocytochemistry, enzyme immunoassays etc. Most of these assays are either non-quantitative or are population studies of protein expression. Due to the heterogeneity of tissues & cells, there is now an emerging trend towards single cell analysis.

Flow cytometry has been around for decades and is a quantitative way of studying protein expression in single cells. However, current flow cytometers are huge, complex and often requires a dedicated operator with many years of experience.

Millipore, in partnership with Guava, is now offering to researchers all the advantages of flow cytometry in a compact & simple to operate platform. Millipore is also developing a full range of validated assay kits for stem cells, apoptosis & cell signaling research.

This talk will focus on the importance of single cell analysis and how the Guava system can speed up the workflow in various fields of biological science.

1.The Guava® ViaCount® Reagent provides absolute cell count and viability data on cell suspensions from a variety of cultured mammalian cell lines. The Guava ViaCount assay distinguishes between viable and non-viable cells based on the differential permeability of DNA-binding dyes in the ViaCount Reagent. The Guava ViaCount assay is intended as a rapid and reliable method for determining cell count and viability.

2.The Guava Nexin™ Assay utilizes Annexin V-PE to detect phosphatidylserine on the external membrane of apoptotic cells. The cell impermeant dye, 7-AAD, is included in the assay as an indicator of membrane structural integrity. 3 populations of cells can be distinguished: Non-apoptotic cells, Early apoptotic cells, Late stage apoptotic and Dead cells.

3.The Guava® Cell Cycle Reagent contains the Propidium Iodide dye and is used to determine the percentage of cells in G0/G1, S and G2/M phases based on DNA content.
